US healthcare company Johnson & Johnson (NYSE: JNJ) on Friday announced the release of two-year Phase 3 results from ICONIC-TOTAL assessing ICOTYDE (icotrokinra) in adults and adolescents with moderate high-impact plaque psoriasis.
The oral peptide, which blocks the IL-23 receptor, was found to deliver sustained skin clearance through to Week 112.
Clear or almost clear skin (Investigator's Global Assessment 0/1) increased from 57% at Week 16 to 70% at Week 112 among treated patients. Site-specific complete clearance at Week 112 reached 60% in scalp psoriasis, 89% in genital psoriasis and 63% in psoriasis affecting the hands and/or feet.
Nail psoriasis also improved, with mean modified Nail Psoriasis Severity Index (mNAPSI) scores rising from 33% at Week 16 to 71% at Week 112. Safety findings remained consistent with the established profile, with no new signals reported.
ICONIC-TOTAL is part of a broader Phase 3 programme evaluating ICOTYDE across plaque psoriasis and related inflammatory conditions, including studies comparing the drug with placebo and deucravacitinib. ICOTYDE is approved in multiple regions for moderate-to-severe plaque psoriasis and is being studied in psoriatic arthritis, ulcerative colitis and Crohn's disease.
